About the Job

Summary of Position

This individual will conduct reverse genetic experiments utilizing CRISPR/Cas9-mediated gene editing technology. The bulk of the project will consist of generating a set of reagents (genetically-modified human cell lines) and then characterizing them for deficits in genetic recombination and telomere maintenance. This individual should have previous experience working in a molecular biology laboratory and to especially have experience in performing human tissue culture. Familiarity with other molecular techniques such as PCR, gel electrophoresis, DNA cloning, etc. is highly desirable.

Essential and Other Functions:

50% Perform experiments related to utilizing CRISPR/Cas9 to generate genetically modified human cell lines, Use CRISPR/Cas9 to generate null and conditionally-null cell lines for genes related to Holiday Junction resolution.

30% Perform and design experiments related to characterizing genetically modified human cell lines.
Use a bevy of molecular assays (T-SCE, gene targeting, HDR reporter assays, etc.) to elicit whether any of the genetically modified cell lines are defective for HJ resolution and if so, where precisely are they defective.

10% General laboratory responsibilities
Help out with general laboratory well-being, ordering, cleaning, organizing, etc.

10% Supervision of student employees
Oversee all of the undergraduate employees who are used to wash glassware and generally help with the up keep of the laboratory.

Qualifications

All required qualifications must be documented on application materials

Required Qualifications:
BA/BS plus at least one year of experience using CRISPR/Cas9 to perform gene editing in human cells, as well as working with human tissue culture and with basic molecular biological techniques, or a combination of related education and experience totaling at least five years.

Preferred Qualifications:
Someone with a working knowledge of HJ resolvases and the biology associated with genetic recombination.
